The main source of estrogens in a normally adult female cycling is the ovarian follicle, which releases 70 to 500 μg of estradiol daily, depending on the phase of the menstrual cycle. After menopause, the most endogenous estrogens are obtained by converting Androstenedione, secret by the adrenal cortex, to estrone in peripheral tissues. Thus, estrone and sulfate conjugated form, estrone sulfate, are the most common circulating estrogens in postmenopausal women.

Estrogens act through binding to the nuclear receptors in the estrogen of a flexible tissue. To date, two estrogen receptors have been identified. They differ in the proportion of fabric tissue.

Circulating estrogens modulate the secretion of the pituitary gland gonadotropins, luteinizing hormone (LH) and follicle stimulating hormone (FSH), through the mechanism of negative feedback. Estrogens take measures to reduce elevated levels of these gonadotropins in postmenopausal women.

  • Signs of blood clots such as pain, swelling and redness of the legs, sudden chest pain, and difficulty in breathing
  • Signs of liver problems (such as yellowing of the skin or the whites of the eyes (jaundice))
  • Signs of high blood pressure (such as headache, tiredness, dizziness)
  • Migraine-like headache happening for the first time
  • Signs of allergic reaction (such as skin rash, itching, shortness of breath, difficulty in breathing, and a swollen face)
  • Inflammation of the colon (part of the intestine) which may present as lower left sided abdominal pain and/or bloody diarrhea
  • Growth of benign meningioma (a tumor of the membranes around the brain or spinal cord)
  • Signs of breast cancer (breast pain or tenderness, swollen breasts, discharge from the nipples and changes in breast tissue)

HOW TO MANAGE SIDE EFFECTS

Nausea and Vomiting:

Take PREMARIN 0.625MG with or just after meals. Stick to simple meals. Avoid eating oil rich or spicy foods. Consult your doctor if your nausea worsens.

Headache:  

Rest and relax. Keep yourself hydrated by drinking plenty of fluids such as water or electrolytes. Applying a pain-relieving balm on your forehead can be beneficial. Do not consume excessive alcohol, as it could aggravate your headache. Consult your doctor if your headache worsens.

Dizziness:

PREGNANCY

PREMARIN 0.625MG is not intended for use in pregnant women. Stop taking PREMARIN 0.625MG and consult your doctor immediately if you accidentally become pregnant while taking PREMARIN 0.625MG.

BREASTFEEDING

DRIVING AND USING MACHINES

PREMARIN 0.625MG is unlikely to affect your ability to drive or to use machines. However, do not drive or operate any heavy tools or machines if your ability is affected by PREMARIN 0.625MG.

ALCOHOL

Avoid consumption of alcohol while taking PREMARIN 0.625MG, as it may increase the risk of side effects.

KIDNEY

LIVER

ALLERGY

LUNGS

HEART DISEASE

OTHERS

PREMARIN 0.625MG is not recommended for use if you:

  • Have cancer in the uterus lining (endometrium), or if you are suspected of having it
  • Have any unexplained vaginal bleeding
  • Have untreated endometrial hyperplasia (excessive thickening of the layer of uterus)
  • Have or had blood clot in the veins (thrombosis, deep venous thrombosis, pulmonary embolism)
  • Have a blood clotting disorder (such as Protein C, Protein S, antithrombin deficiency)
  • Have porphyria (inherited disorder affecting RBC)

A. Drug-Drug Interaction:
